The competition will take place at 35 sports facilities in various districts of Rio de Janeiro. Most of the Olympic venues will be located in the Barra da Tijuca area. It will host competitions in cycling, gymnastics, trampoline, tennis, water sports, basketball, judo, taekwondo, golf, badminton, table tennis and weightlifting. The rest of the facilities will be located in three zones: Copacabana, Maracana and Deodoro. In addition to Rio de Janeiro, football matches will take place in El Salvador, Sao Paulo, Belo Horizonte and Brasilia.

The readiness of the Olympic facilities is a concern of the International Olympic Committee. On May 9, 2014, IOC Vice President John Coates announced that the construction of the Olympic infrastructure was far behind schedule. In May, preliminary consultations began regarding the transfer of the Olympic Games to another city. London, Moscow and Glasgow were considered as cities for the transfer of the Games. At the 2016 Summer Olympics, competitions will be held in 41 disciplines in 28 sports. The Games program includes two new sports: golf and rugby. According to preliminary estimates, 12,500 athletes from 205 countries will participate in the medal competitions.
